Serial crystallography for the masses?
نویسنده
چکیده
Serial crystallography should be possible with a much wider range of radiation sources as Ayyer et al. [IUCrJ (2015), 2, 29-34] show that crystallographic intensities can be recovered from randomly oriented frames which are too sparse for indexing.

متن کاملTime-resolved structural studies with serial crystallography: A new light on retinal proteins
Structural information of the different conformational states of the two prototypical light-sensitive membrane proteins, bacteriorhodopsin and rhodopsin, has been obtained in the past by X-ray cryo-crystallography and cryo-electron microscopy. However, these methods do not allow for the structure determination of most intermediate conformations. Recently, the potential of X-Ray Free Electron La...
متن کاملSerial crystallography at synchrotrons and X-ray lasers
Serial crystallography was developed for the use at free-electron lasers but the approach has recently also been adapted to synchrotron sources. Here we discuss how the synergy between the two X-ray sources will facilitate a wide application of the technique in microcrystallography, room-temperature structure determination and time-resolved studies.
متن کاملDevelopment of a dose-limiting data collection strategy for serial synchrotron rotation crystallography
Serial crystallography, in which single-shot diffraction images are collected, has great potential for protein microcrystallography. Although serial femtosecond crystallography (SFX) has been successfully demonstrated, limited beam time prevents its routine use. Inspired by SFX, serial synchrotron crystallography (SSX) has been investigated at synchrotron macromolecular crystallography beamline...
متن کاملRadiation Damage in XFEL: Case study from the oxygen-evolving complex of Photosystem II
Structural changes induced by radiation damage in X-ray crystallography hinder the ability to understand the structure/function relationship in chemical reactions. Serial femtosecond crystallography overcomes this problem by exposing the sample to very short and intense laser pulse leading to measurement before destruction. Here we use molecular modeling to map the radiation damage during the 1...
